Is the frozen squash the only ingredient different from making this soup before? Could bitterness come from another ingredient? If you still have some of the frozen squash, cook a little bit in its own and taste.
Further notes. Frozen produce rarely goes bad, but if your tested sample is bad, either toss it and/or take to store for replacement. If the squash tasted ok, think what else in the recipe could have given butter taste to the soup.
