making peaches and cream cake on cup of Jo, can I use Greek Yogurt and should I add any sugar?

I had my eye on this one too. I think Greek yogurt would be fine. If you read the comments on the website, some felt it wasn't sweet enough. You might want to add sugar to your personal taste.
