You mean, instead of the white wine?
Most DIY vermouth recipes use white wine, I think to let the herbal flavors show.
On the other hand, this one already has port and brandy, so if the flavors can stand up to them, it's probably worth a try.
PS If you go with the red wine, I would use same volume as recipe calls for the white wine.
