I have a 500 gr can of unsweetened chestnut puree I would like to use up. Should I use the whole can?

Misinterpreted your question.
Yes, in soups and stews, like this one, you can use non-exacflt measures of ingredients, depending on what you have on hand.
If you like, go ahead and use all the 500g contents vs 400g called for in the recipe.
Ir, if you gave another use for the puree in a few days, save the 100g for that.

This is a versatile and delicious starch/vegetable, with lots of ways to eat it.

If the label on the can of chestnut puree has some time to run, and you can get fresh chestnuts near you, maybe use (a smaller amount of) those for the soup and keep the can for another time.
If you must or want to use the 500g can now, yes open it. One source says it lasts 4-5 days in fridge and 9-12 months in freezer, so you could use some now and freeze the rest for later.

If you do open the whole can but don't need all of it for the butternut squash-miso soup recipe, there are other ways to use it up.

SWEETENED VERSION OF THE PUREE:
make Mont Blanc dessert with whipped cream
on toast, like jam or other sweet spreads
with fruit and/or ice cream for dessert
baked in cakes
used as topping or icing for cakes, meringue desserts, etc

UNSWEETENED VERSION OF PUREE:
in other soups
in stuffing (dressing) for turkey or chicken
with red cabbage and/or root vegetables as a side dish.
